Descrição

Environmental DNA sequence data of invasive species along the harbors in the northern regions of Vastra Gotaland. Sequence data was obtained from both water samples, and plankton samples of two sampling events, one in the summer and one in the autumn of 2024. The amplicons were amplified by COI Leray-XT primers: Wangensteen et al. 2018 and annotated against an inhouse database of invasive and non-indigenous species of Norway, Sweden and EEA created with EchoPipe: Stensrud et al. in press. The data was created for Länsstyrelsen Vastra Gotaland and financed through HaV.

This dataset was published via the SBDI ASV portal.

Plankton samples were collected using an Apstein 50 plankton net with 100 μm mesh size (Hydro-Bios, Germany), by lowering the net 1-2 meters above the bottom and retrieving it at ~1 meter per second. This was done 2-3 times per sampling site until enough plankton material had been collected. The collected material was then transferred to a plankton sieve with a mesh size of 90 μm and cleaned with local seawater before being transferred to a 15 ml sterile Falcon tube. To preserve the samples, the Falcon tube was filled with 99% ethanol and stored in the refrigerator until DNA extraction. The water samples were collected using a Ruttner collector (1.7 L) where water masses from two sites were pooled. For each site, water was collected from ~1-2 meters above the bottom and surface water and mixed in a bucket that was previously sterilized with 10% chlorine, washed with ethanol and rinsed three times with local seawater before each sampling. New gloves and a new sterile 60 ml syringe were used at each site to filter between 1140 and 1800 ml of water through a PureFlo disk filter (GF/PES 5.0/0.8 μm, Saint-Gobain Filter, USA) to capture environmental DNA. The filters were then immediately frozen in a Dry Shipper cooled with liquid nitrogen (-196°C) and then stored at -20°C when returned to the laboratory until DNA extraction.

Introduction Invasive and non‑indigenous species are one of the biggest challenges to biodiversity and can have major ecological and economic impacts. The number of Invasive and non‑indigenous species has increased recently due to factors such as global warming and increased connectivity, including as a result of shipping. Early detection of non‑indigenous and potential invasive species is important so that effective measures can be taken in time to prevent their establishment. Five different harbors in the northern parts of Västra Götaland's west coast have been surveyed with environmental DNA to detect invasive and non‑indigenous species during the summer and autumn of 2024. The survey was conducted by WaterCircle AB in collaboration with eDNA solutions AB on behalf of the Länsstyrelsen of Västra Götaland. The environmental DNA was amplified with COI Leray-XT primers and annotated against an internal database of invasive and invasive species from the EU, Norway and Sweden. A total of 36 non‑indigenous species and five additional non‑indigenous species groups were detected. Of these non‑indigenous species, five had not previously been recorded in Sweden and one additional species had only previously been detected using environmental DNA. Of the 41 non‑indigenous species/species groups, 20 were considered to be at least potentially high risk of being invasive according to the EEA, Norwegian and/or Swedish risk classification, of which nine were at least potentially high risk according to the Swedish Species Register (Artdatabanken). The largest harbors Strömstad and Preemraff Brofjorden had the most unique detection of both non‑indigenous and invasive species, which supports that these ports in particular are hotspots for the introduction of new invasive species, which can then spread further.
