Description
This project reports genetic data from the non-indigenous species (NIS) monitoring program in Sweden in 2024, as commissioned by the Swedish Agency for Marine and Water Management, SwAM. The data were collected and submitted by Seanalytics, www.seanalytics.se in collaboration with University of Gothenburg. Sampling locations were situated in ports and marinas along the Swedish coast, including the Bothnian gulf, the Swedish West coast and Southern coast. Three types of samples (water samples, plankton samples, hard bottom samples) were collected during two seasons - late spring and late summer. Species were identified using COI metabarcoding. The dataset includes ASVs (amplicon sequence variants) and their associated metadata. Méthodes d'échantillonnage
All sampling was carried out by Seanalytics. Plankton was collected with a Hydro-Bios Apstein 90µm net, diameter 40 cm opening. Samples were pulled from the bottom upwards, two pulls per site. The samples were collected in a 1-litre jar and fixed with 96% ethanol in an amount that ensured that the final concentration did not fall below 70%. A total of three samples were collected per site. Water samples were taken with Ruttner water sampler, 1 liter at the surface and 1 liter at a depth of 3-6 meters at each location. These samples were combined in a 2-liter disposable container and stored in cool bags with ice packs until filtration. A total of 10 samples from each location were collected. Water samples were filtered (Sterivex 0.45µl) immediately with a peristaltic pump connected to an electric drill and fixed with 95% ethanol and sealed with stoppers. Between rooms, 10% chlorine solution was pumped through and then rinsed thoroughly. This was also done before the blank sample (0.5 liters of deionized water) for contamination control. All samples were stored in a -20° C freezer until extraction.

Objet | Samples of water (eDNA), plankton and settlement plates were taken in ports and marinas along the Swedish coastline during early summer and autumn 2024. A total of 249 samples were taken (including blanks and positive controls), leading to identification of 494 species based on 4264 amplicon sequence variants (ASVs). Scanning for non-indigenous species (NIS) resulted in 873 observations of 33 known and putative NIS as well as 10 crytogenic species. Raw genetic sequence data are submitted to ENA. |
